Alcohol

· Severity: Major

· Notes for Consumers: Do not drink alcohol while taking this medication. Drinking alcohol while taking this medication can cause serious side effects, such as central nervous system (CNS) depression, and can increase the risk of falling. Call emergency services right away if you experience symptoms of CNS depression: slow or shallow breathing, shortness of breath, feeling faint, dizziness, confusion, trouble staying awake.

· Notes for Professionals: Advise patients to avoid the use of alcohol with daridorexant as concurrent use increases the risk for additive CNS depression and impairment.

Grapefruit juice

· Severity: Major

· Notes for Consumers: Avoid grapefruit juice and grapefruit while taking this medicine. Serious side effects such as slurred speech, confusion, excess drowsiness, severe weakness, or difficulty breathing may occur. Contact your care team right away if you have any of these side effects. Do not drive, operate machinery, or do anything that requires mental alertness after taking your nighttime dose of Daridorexant.

· Notes for Professionals: Avoid concomitant use of daridorexant and grapefruit juice. Concomitant use may increase daridorexant exposure and the risk for daridorexant-related adverse effects. Daridorexant is a CYP3A substrate and grapefruit juice is a strong CYP3A inhibitor. Concomitant use of another strong CYP3A inhibitor increased daridorexant overall exposure by over 400%.
