· Severity: Major

· Notes for Consumers: Side effects from Enfortumab Vedotin may get worse if you drink Grapefruit Juice, including nausea, vomiting, mouth sores, and pain, tingling, numbness in the hands or feet. Discuss any new or worsening side effects with your health care provider. Contact your health care provider immediately if you notice fever or chills, sore throat, bruising or pinpoint red spots on the skin.

· Notes for Professionals: Advise patients to avoid grapefruit juice while taking enfortumab vedotin due to a likely increased risk of exposure to unconjugated monomethyl auristatin E (MMAE), which may increase the incidence or severity of enfortumab-vedotin toxicities. MMAE, the microtubule-disrupting component of enfortumab vedotin, is a CYP3A4 and P-gp substrate; grapefruit juice is a dual P-gp/strong CYP3A4 inhibitor.
