Food

· Severity: Severe

· Notes for Consumers: Nilotinib needs to be taken at least 1 hour before a meal or 2 hours after a meal. Do not take Nilotinib with food. Taking Nilotinib with food may increase side effects. Tell your doctor about any unusual effects, such as irregular or fast heartbeat, chest pain, palpitations, or unusual fatigue, weakness or fever.

· Notes for Professionals: Do not administer Nilotinib with food. Nilotinib needs to be taken at least 1 hour before a meal or 2 hours after a meal. Food increases the bioavailability of nilotinib. Increased nilotinib serum concentrations may occur if the drug is taken with food, possibly resulting in QT porlongation, palpitations, torsade de pointes (TdP) and/or myelotoxicity.

Grapefruit juice

· Severity: Major

· Notes for Consumers: Do not take Nilotinib with grapefruit, grapefruit juice, or supplements that contain grapefruit extract. Taking Nilotinib with grapefruit or grapefruit juice may increase side effects of Nilotinib. If you accidentally take Nilotinib with grapefruit or grapefruit juice, immediately tell your doctor about any unusual effects.

· Notes for Professionals: Avoid the use of nilotinib with grapefruit, grapefruit juice, or grapefruit products. The AUC value of nilotinib increased by 1.3-fold when nilotinib was administered with a single intake of double-strength grapefruit juice.
